Samurai in Ghost of Tsushima is one of the 4 classes available with Ghost of Tsushima: Legends DLC. Classes can be unlocked after you play the game a little bit. The first class you can unlock is only when your player level reaches Rank 8. This is where you can select the first Class of your choice and equip.

Samurai Information
Samurai players can run straight toward combat and stay there, sustaining their health while chopping enemies down left and right. When you’re overwhelmed, use the Hachiman’s Fury ultimate attack to slash through enemies in a flurry of strikes.
The Samurai resides on the frontlines, crossing swords face-to-face with the opposition. This class deals heavy damage, but it can also endure punishment from hostile onslaughts to a greater degree than the other characters. If you gravitate towards the tank role, you will feel at home with the Samurai.
